摘 要

银杏(Ginkgo biloba)是我国特有的多用途经济树种,属国家二级保护树种。本论文选用南京林业大学校园内的银杏花粉为材料,将其放置在常温、0℃、-20℃以及-70℃的条件下进行贮藏,分别在贮藏0d、7d、14d和21d的时候对花粉的

内含物质包括:可溶性蛋白质(考马斯亮蓝G-250染色法)、可溶性糖(蒽酮比色法)、CAT(紫外吸收法)、POD(愈创木酚法)、SOD(氮蓝四唑还原法)和总黄酮量(分光光度计法)的测定。了解贮藏期间银杏花粉生活力的变化特点,对收集的数据进行分析得出以下结论:

  1. 本次试验中,可溶性糖、可溶性蛋白质的含量遵循先增后减少趋势,在贮藏21d内,银杏花粉的可溶性糖、可溶性蛋白质含量虽然在不断变化,但是始终含量都较高。在贮藏14d下,-70℃的可溶性蛋白和可溶性糖含量最高,其次是-20℃,最后是常温贮藏的花粉。
  2. 黄酮类物质是银杏一种很重要的营养物质,银杏的叶、花粉、果实均含有黄酮类物质。本次试验发现常温、-20℃和-70℃贮藏条件下,总黄酮的含量随着贮藏时间逐渐增加,从而保持花粉的活力。

3.在植物体内活性氧自由基的产生与保护酶系统 SOD、POD及CAT 在本次试验中,-70℃贮藏下的花粉SOD、CAT以及POD的活性均先增加后减少,这是因为受到低温胁迫而启动了酶保护系统。

Study on the Relationship between Storage Conditions and Ginkgo Pollen Internal Nutrients

ABSTRACT

Ginkgo biloba is China's unique multi-purpose economic tree species, is a national secondary protection tree species. In this paper, the ginkgo pollen in the campus of Nanjing Forestry University was selected and stored at room temperature, 0 ℃, -20 ℃ and -70 ℃, and the pollen was stored at 0d, 7d, 14d and 21d respectively. Including soluble protein (Coomassie brilliant blue G-250), soluble sugar (anthrone colorimetric method), CAT (ultraviolet absorption method), POD (guaiacol method), SOD (nitrogen blue tetrazole reduction Method) and total flavonoid content (spectrophotometric method). To understand the characteristics of gingko pollen viability during storage, analyze the collected data and draw the following conclusions:

1. The content of soluble sugar and soluble protein in this experiment followed the trend of decreasing first and then decreasing. During the 21 days storage, the soluble sugar and soluble protein content of Ginkgo pollen were constantly changing, but the content of soluble protein was higher. The soluble protein and soluble sugar content were the highest at -20℃, followed by -20℃, and finally pollen at room temperature.

2. Flavonoids is a very important nutrient ginkgo, ginkgo leaves, pollen, fruit contains flavonoids. In this experiment, it was found that the content of total flavonoids increased with the storage time at room temperature, -20℃ and -70℃, so as to maintain the vitality of pollen.

3.The activity of SOD, POD and CAT in the plants were increased and decreased at the temperature of -70℃, and the activities of SOD, CAT and POD were increased and decreased. Low temperature stress and start the enzyme protection system.
