柳树小热激蛋白Hsp20基因家族的鉴定和表达分析毕业论文

摘 要

小热激蛋白在植物中有最丰富的含量和最广泛的分布,其一直是植物胁迫研究的重点和热点,而小热激蛋白Hsp20在柳树响应多种环境胁迫包括高温等以及在柳树生长发育的过程中具有非常重要的作用,柳树作为一个重要的树种,人们仍对其Hsp20的基因家族缺少全面的研究和了解。

该试验通过柳树小热激蛋白基因家族的鉴定和表达分析的研究,获取柳树的Hsp20基因家族成员和其在不同组织中的表达模式。本论文选用簸箕柳雌株在生长室进行扦插,45天后取其根、茎和展开叶、顶芽和树皮组织,然后提取上述样品组织的RNA,进行转录组测序,分析柳树不同组织细胞中基因的表达情况。

结果发现,一方面可以知道有60个基因家族成员并分为7个亚族,另一方面在簸箕柳的根、茎、叶、顶芽和树皮组织进行转录组测序,发现这些基因家族成员在不同的组织中表达模式不同,并且同一个家族成员在不同组织中表达模式可能相同也可能不同,为其他种类的柳树甚至其他树种的Hsp20研究提供参考。从而促进柳树树种的发展,有利于柳树更好的适应生长发育环境,也有利于柳树更好的作为防风固沙林和速生率产林,提供更大的经济价值和社会价值。

Identification and Expression Analysis of Hsp20 Gene Family of Small Heat Shock Protein in Willow

Abstract

Small heat shock protein is the most abundant protein with the most extensive distribution in plants. It has always been the focus and hot subject for plant stress research. Small heat shock protein Hsp20 plays a very important role in willow’s response to various environmental stresses including high temperature and the process of willow’s growth and development. As an important tree species, willow is still lack of comprehensive research and understanding of its Hsp20 gene family.

In this experiment, through identification and expression analysis of willow’s small heat shock protein gene family, Salix purpurea Hsp20 gene family members and their expression patterns in different tissues were obtained. In this paper, the female plant of Salix suchowensis Cheng was selected for cuttage in the growth chamber. After 45 days, its root, stem, expanded leaf, terminal bud and bark tissues were collected. Then, RNA of the above sample tissues was extracted for transcriptome sequencing. Besides, the gene expression in different tissue cells of Salix was analyzed.

The result showed that, on the one hand, there were 60 gene family members, which were divided into 7 subgroups. On the other hand, via transcriptome sequencing for the root, stem, leaf, terminal bud and bark tissues of Salix suchowensis Cheng, it found that the expression patterns of these gene family members were different in different tissues, and the expression patterns of the same family member in different tissues might be same or different. The research provides reference for Hsp20 research of other willow species and even other tree species. In addition, it promotes the development of willow tree species, which is conducive to better adapting willow to the growth and development environment, as well as better using willow as windbreak and sand fixation forest and fast-growing yield forest to provide greater economic value and social value.
